Plasma was tested by direct ELISA for binding to either mixed mold or recombinant human cochlin at all plasma dilutions, and the presence of bound antibody was determined using a peroxidase conjugated-goat anti-human IgG heavy and light chain antibodies . Detection was performed by treatment with H2O2 in ABTS substrate , and stopped after 30 min by adding ABTS Peroxidase Stop Solution containing 5% SDS. Change in absorbance was read at 405 nm in an ELISA reader equipped with Biolinx 2.2 software . IgG Concentrations in plasma were determined by coating purified Human IgG

At the end of all incubations, PBMC were centrifuged, RNA harvested and supernatants were collected and stored in â20 °C.
